RESUMO

Clove plant (Syzygium aromaticum) is one of the Myrtaceae family. It's a common flavor in food and the traditional medicine. The study's objective was to ascertain whether the clove bud aqueous extract (CAE) and CAE + nanosilver have any biological effects on immune cells and HT-29 colon cancer cell line. Nanosilver was produced through green synthesis approach using CAE. Produced nanosilver was characterized via electron microscope (scanning, SEM) and ultraviolet-visible spectroscopy. CAE and CAE + nanosilver were examined for their active biomolecules using FTIR analysis, p53 contents using real-time PCR, apoptosis and cell cycle arrest power on HT-29 cancer cell line via flow cytometerty and immunomodulatory potential utilizing MTT assay. Results cleared that a spherical nanosilver with a diameter range of 53 nm was formed by CAE. There were several active biomolecules in CAE and CAE + nanosilver. CAE and CAE + nanosilver increased the p53 protein expression and apoptotic cell number in HT-29 colon cancer cells. CAE and CAE + nanosilver could arrest HT-29 cells at the phase G2/M. CAE and CAE + nanosilver stimulated quiescent and PHA-pre-treated splenic cells at higher concentrations, and CAE suppressed quiescent splenic cell when diluted. In conclusion, the safe edible Syzygium aromaticum plant can be utilized to make anti-tumor agent, essentially for colon tumor. As Syzygium aromaticum plant could stimulate immune cells, it can be used as immune-stimulatory agent that can help fight tumor and tumor development.

BACKGROUND: Much published data exists on the position of cervicothoracic ganglion, but a little published research has been done on the cervicothoracic system of dog. Herein, we illustrated topographical position and shape of each ganglion of cervicothoracic system to determine the distribution of nerves dispersing from them on two sides, left and right. MATERIALS AND METHODS: Our work designed on the usage of 10 healthy adult dogs. Left cervicothoracic sympathetic system is represented by two ganglia: caudal and middle ganglion, while the right system is represented by three ganglia: caudal, middle cervical and small accessory ganglia. RESULTS: Left caudal cervical ganglion was elongated triangular, while the right one was elongated spindle in shape. Left caudal cervical ganglion was located on lateral surface of longus colli muscle, at the first intercostal space, while the right one was located at the level of the second rib. Left middle cervical ganglion was ovoid in shape and located at the first intercostal space, while the right one was located at the level of the second rib. There were two nerve trunks forming ansa subclavian trunk on both sides. There were three sympathetic-parasympathetic communicating branches on both sides. CONCLUSIONS: Our study recorded the first observation of left pericardial branch in dog, which originated from the caudal angle of middle cervical ganglion. There was a small ganglion located on the lateral surface of trachea at the level of the first rib.

BACKGROUND: This work was designed to provide a morphologic, morphometric and histochemical description of the eye of the African straw-coloured fruit bat (Eidolon helvum). An explanation of the optical role of the choroidal papillae in the vision of megachiropteran bats was provided. MATERIALS AND METHODS: Enucleated eyes of captured fruit bats were measured and processed for light microscopy. RESULTS: Typical gross features of the mammalian eye including an anterior transparent cornea, posterior whitish sclera and a golden-brown iris surrounding a round pupil were observed in the eye. Presence of undulating retina typically found in megachiropterans was also seen. The ratio of mean corneal diameter to mean axial eye diameter was 0.58 ± 0.08. The histochemical investigation of the eye indicated the presence of mucins, proteoglycans, hyaluronic acid, glycogen and/or glycoproteins in the corneal, scleral, choroidal and retinal tissues. CONCLUSIONS: The presence of reflective materials of the tapetum lucidum on the undulating retina was shown to be a morphological adaptation for increased light sensitivity as each parabolic surface of the choroidal papillae served as a convex mirror, reflecting the light rays to the adjacent parabolic surface, thus sensitising photoreceptors in affected regions. This phenomenon thus empowers megachiropteran bats with improved scotopic visual capability and could explain why most of them are reliant on their vison without the need for echolocation.

BACKGROUND: The current study was designed to perform a transmission electron microscopic investigation focusing on the dark and light pancreatic acinar ß-cells of young domesticated pig (Sus Suidae). MATERIALS AND METHODS: This study depended on the fresh pancreatic specimens from 5 healthy young (2-month-old) pigs that were collected immediately after they were slaughtered at the abattoir of Abdelkader Alexandria, Egypt. RESULTS AND CONCLUSIONS: In our findings, the acinar pancreas was formed of pyramidal pancreatic acinar cells with large spherical nuclei of condensed heterochromatin at the periphery and prominent eccentric nucleoli. Zymogen granules were observed at the apical region of the acinar cells, and they appear as electron dense bodies. Numerous mitochondria and Golgi complexes observed in the acinar cell cytoplasm. The electron dense acinar cells were joined by junctional complexes. The rough endoplasmic reticulum was more prominent in the electron-dense acinar cells than did electron-lucent acinar cells. There was no connective tissue capsule separate the acinar portion of pancreas from the pancreatic islets. The pancreatic islets mainly formed of ß-cells. The irregular α-cells possess numerous small granules. The cytoplasmic ß-cells granules were surrounded by hallow area and enclosed by a limiting membrane. Delta cells were generally polygonal in shape and found in clumps throughout the islet and they were also identified in between ß-cells. Their granules were of moderate electron density and were generally smaller than ß-cells' granules. The limiting membrane was tightly enclosed the delta cells granules and the hallow area around the granule were found similar to the granules of ß-cells.

In this research, some plant seeds powder was evaluated to find their potential effect to rule diseases of food poisoning. Antimicrobial effect of five plant seeds was examined contra Bacillus cereus, Staphylococcus aureus, Escherichia coli, Klebsiella. pneumonia and Candida albicans by using well diffusion method. Antimicrobial activity studies revealed high potential activity of plant seeds powder of Nigella sativa L., cucurbita pepo, Sesamum radiatum, Trigonella foenum-graecum, Linum usitatissimum with variable efficiency contra tested microbial strains with concentration of 100 mg/ml, except Sesamum radiatum scored no effect. The T. foenum and N. sativa seed powder showed the largest inhibition zone (24-20 mm) contra K. pneumonia, followed by S. aureus (20-18 mm) and C. albicans (15mm) respectively. The five plant seeds powder exhibited bacteriostatic and bactericidal effects with MIC's 20 and MBC 40 mg/ml against K. pneumonia, and MIC's 40 and MBC 60 mg/ml against S. aureus. The results of this study indicated that plants seeds powder have promising antimicrobial activities and their potential applications in food process. It could be utilized as a natural medicinal alternative instead of chemical substance.


Nesta pesquisa, o pó de sementes de plantas foi avaliado para encontrar seu efeito potencial no controle de doenças de intoxicação alimentar. O efeito antimicrobiano de cinco sementes de plantas foi examinado contra Bacillus cereus, Staphylococcus aureus, Escherichia coli, Klebsiella pneumonia e Candida albicans usando o método de difusão bem. Estudos de atividade antimicrobiana revelaram alto potencial de atividade de sementes de plantas em pó de Nigella sativa L., Cucurbita pepo, Sesamum radiatum, Trigonella foenum-graecum, Linum usitatissimum com eficiência variável contra cepas microbianas testadas com concentração de 100 mg / ml, exceto Sesamum radiatum com pontuação não efeito. O pó de sementes de T. foenum e N. sativa apresentou a maior zona de inibição (24-20 mm) contra K. pneumonia, seguido por S. aureus (20-18 mm) e C. albicans (15 mm), respectivamente. Members of the family Diclidophoridae are potentially dangerous species for the puffer fish aquaculture worldwide. They are parasitic polyopisthocotyleans, with a posterior haptor equipped with clamps for attachment to the host's surface, allowing the worm to resist the flow of water to maintain its position on gills. The anterior body of the worm is deformable, allows the worm to feed on blood sucked from fish gills. The present study is the first description of a Heterobothrium species from the gills of the tiger puffer Lagocephalus sceleratus (Tetraodontidae) from the coasts of the Arabian Gulf at Jubail, Saudi Arabia morphologically by light microscopy as well as by molecular analysis of the parasite partial 28S rRNA through multiple sequence alignments and phylogeny by maximum likelihood analysis which is provided for the first time for the described species. Seventeen tiger puffer fish were captured alive from marine water off Saudi Arabia; gills were separated and further examined for parasitic infection. Nine fish were found infected with a monogenean parasite which was robust, equipped by two buccal organs at the tapered anterior end; the posterior haptor was rectangular with four symmetrically arranged clamps, with no isthmus. Marginal hooks absent. Ovary elongated, U-shaped, testes numerous, irregularly shaped and extended from the posterior part of the ovary to the anterior margin of the haptor. Copulatory organ muscular, as a spherical cup armed with 12 to 15 genital hooks. The molecular analysis of the parasite 28s rRNA and phylogeny revealed a percentage of identities between 87.47-89.09%, with Diclidophoridae species within the monophyletic clade of Mazocraeidea where a maximum percentage of 89.09% were obtained for the morphologically different sister taxon H. okamotoi. The results obtained from molecular analysis are consistent with the conclusions drawn from morphological classification where that the parasite recorded was morphologically similar to H. lamothei which was not characterized by molecular analysis before. The recovered sequences were deposited into the GeneBank under accession number MT322610.(AU)


Os membros da família Diclidophoridae são espécies potencialmente perigosas para a aquicultura de peixes puffer em todo o mundo. Eles são parasitas poliopisthocotyleans, com uma hélice posterior equipada com pinças para fixação na superfície do hospedeiro, permitindo que o verme resista ao fluxo de água para manter sua posição nas brânquias. O corpo anterior do verme é deformável, e permite que o verme se alimente de sangue sugado das guelras dos peixes. O presente estudo é a primeira descrição de uma espécie Heterobothrium das guelras do tigre Lagocephalus sceleratus (Tetraodontidae) das costas do Golfo Arábico em Jubail, Arábia Saudita, usando morfologia por microscopia leve, bem como análise molecular do rRNA parcial do parasita 28S através de alinhamentos de sequências múltiplas e filogenia por análise de máxima verossimilhança que é fornecida pela primeira vez para as espécies descritas. Dezessete peixes tigre puffer foram capturados vivos da água marinha da Arábia Saudita; as brânquias foram separadas e mais tarde examinadas para detecção de infecção parasitária. Nove peixes foram encontrados infectados por um parasita monogênio robusto, equipado por dois órgãos bucais na extremidade anterior afilada; o hortetor posterior era retangular com quatro pinças dispostas simetricamente, sem istmo. Ausência de ganchos marginais. Ovário alongado, em forma de U, testículos numerosos, de forma irregular e estendido desde a parte posterior do ovário até a margem anterior do hortelino. Órgão copulatório muscular, como um copo esférico armado com 12 a 15 ganchos genitais. A análise molecular do parasita 28s rRNA e filogenia revelou uma porcentagem de identidades entre 87,47-89,09%, com espécies Diclidophoridae dentro do clade monofilético de Mazocraeidea onde uma porcentagem máxima de 89,09% foi obtida para o táxon-irmão morfologicamente diferente H. okamotoi. Os resultados obtidos da análise molecular são consistentes com as conclusões tiradas da classificação morfológica onde o parasita registrado era morfologicamente semelhante ao H. lamothei que não era caracterizado pela análise molecular antes. As sequências recuperadas foram depositadas no GeneBank sob o número de acesso MT322610.(AU)

The consumption of inadequately thermally treated fish is a public health risk due to the possible propagation of Anisakis larvae and their antigenic proteins, the causative agent of the zoonotic disease anisakidosis. The present study demonstrated the physiological and histopathological changes that accompanied an oral inoculation of crude extracts from fresh and thermally treated Anisakis Type II (L3) in Wistar albino rats. Nematode worms were isolated from the marine fish Dicentrarchus labrax. They were examined and taxonomically identified using light and scanning electron microscopy. The study was performed in 6 rat groups: a control group (I), a garlic oil (GO) inoculated group (II), a fresh L3 inoculated group (III), a thermally treated L3 inoculated group (IV), a fresh L3 + GO inoculated group (V), and a thermally treated L3 + GO inoculated group (VI). It was observed that rats inoculated with fresh and thermally treated L3 crude extracts showed abnormal oxidative stress markers associated with the destruction of normal architecture of spleen and thymus. GO produced a protective effect in rat groups inoculated with L3 extracts + GO administration via the amelioration of oxidative stress markers, which was confirmed by the marked normal structure of the organs' histology. Cooking of L3 infected fish induced severe physiological and histopathological alterations compared to uncooked infected fish. The administration of garlic before and after fish eating is recommended to avoid the dangerous effect of anisakids, even if they are cooked.(AU)


O consumo de peixes tratados termicamente de forma inadequada é um risco à saúde pública devido à possível propagação das larvas de Anisakis e suas proteínas antigênicas, o agente causador da doença zoonótica anisakidose. O presente estudo demonstrou as alterações fisiológicas e histopatológicas que acompanharam a inoculação oral de extratos brutos de Anisakis Tipo II (L3) frescos e termicamente tratados em ratos Wistar albinos. Vermes nematoides foram isolados do peixe marinho Dicentrarchus labrax e foram examinados e identificados taxonomicamente usando microscopia óptica e eletrônica de varredura. O estudo foi realizado em 6 grupos de ratos: grupo controle (I), grupo inoculado com óleo de alho (GO) (II), grupo inoculado com L3 fresco (III), grupo inoculado com L3 tratado termicamente (IV), grupo inoculado com L3 + GO fresco (V), e grupo inoculado com L3 + GO tratado termicamente (VI). Observou-se que ratos inoculados com extrato bruto L3 fresco e tratado termicamente mostraram marcadores de estresse oxidativo anormais associados à destruição da estrutura normal do baço e do timo. GO produziu um efeito protetor em grupos de ratos inoculados com extrato L3 + administração de GO através da melhoria dos marcadores de estresse oxidativo, que foi confirmada pela marcante estrutura normal da histologia dos órgãos. O cozimento de peixes infectados com L3 induziu alterações fisiológicas e histopatológicas graves quando comparado com peixes infectados não cozidos. Recomenda-se a administração de alho antes e depois da ingestão do peixe para evitar o efeito perigoso dos anisakídeos, mesmo se cozidos.(AU)

BACKGROUND: The available data is scanty about Egyptian water buffalo lips, cheeks, and palate. AIMS: The current investigation was focused on describing the morphology of the lip, cheek, and palate. METHODS: Our study included the gross, light, and electron microscopic examinations of ten heads of the Egyptian water buffalos. RESULTS: The nasolabial plate surface carried numerous scales of keratinized epithelium. Internal labial surface and labial mucocutaneous junctions were covered with stratified squamous keratinized epithelium. Two types of hair follicles in the dermis included ordinary and cavernous types characterized by cavernous space. The conical papillae on the internal aspect of the oral commissure were projected from the mucous membrane. Seromucous glands were occasionally observed under the oral mucous membrane of the commissure and gave positive PAS and AB. Conical papillae density on the inner cheek surface had some variations: the rostral part had large papillae, while the dorsal part had numerous papillae than the ventral part, the caudal part had a smaller number of papillae, while the middle part was devoid of papillae. Parotid duct opening in the buccal vestibule was without papillae. Conical papillae had two surfaces; the rostral surface was highly keratinized than the caudal one. The buccal gland was a compound tubuloacinar mixed (mucoserous) gland and mucus acini only reacted to PAS and AB. The oral surface of palatine rugae was covered with highly keratinized epithelium than the aboral surface. Palatine glands showed PAS and AB positive. CONCLUSION: The result describes the relationship between the available food particles, environmental conditions and the lip, cheek, and palate appearance, and structure.

Parapharyngodon (Oxyurida) is a lizard gastrointestinal nematode parasite with a life cycle including lizards as main hosts. However, some species are known to parasitize anurans. In the present study, P. japonicus isolated from the large intestine of the Egyptian changeable lizard, Agama mutabilis was described and illustrated. Forty five specimens of these animals were collected from south Sinai desert, Egypt during the period from May to September 2017. After necropsy, the body was opened by a longitudinal incision from vent to throat, and the gastrointestinal tract was removed. The esophagus, stomach, small and large intestines were examined separately for helminthes. The recovered nematodes were examined by light and scanning electron microscopy. Thirty six specimens (80.0 %) were found to be naturally infected. The parasite was robust with prominent cuticular transverse annulations. Mouth surrounded by three bilobed lips, each with tiny labial papillae. Three pairs of caudal papillae were observed in male worms; 1 pair precloacal, 1 pair sublateral in cloacal opening line, 1 pair in proximal region of caudal appendage on its narrowed point. The posterior extremity beard dorsally directed caudal appendages. Females were with a conical posterior end terminated at a terminal spike. Ovaries reached esophageal isthmus but not wrapped around corpus. The parasite recorded was compared morphologically and morphometrically with the most similar species, it was found that it was most similar to P. japonicus with new host and locality records.

In an aquatic environment, there is a profound and inverse relationship between environmental quality and disease status of fish. Parasites are one of the most serious limiting factors in aquaculture. Therefore, the present investigation was carried out during the period of February-December 2014 to determine the parasitic infections in the African sharptooth catfish Clarias gariepinus, relative to the capability of internal parasites to accumulate heavy metals. Up to 100 catfish were examined for gastrointestinal helminths and 38% of fish were found to be infected with the cestode Polyonchobothrium clarias. The morphology of this parasite species, based on light and scanning electron microscopy, revealed that the adult worm was characterized by a rectangular scolex measuring 0.43-0.58 (0.49 ± 0.1) mm long and 0.15-0.21 (0.19 ± 0.1) mm wide, with a flat to slightly raised rostellum armed with a crown with two semicircles each bearing 13-15 hooks, followed by immature, mature and gravid proglottids which were about 29-55 (45), 16-30 (24) and 15-39 (28) in number, respectively. The mature proglottid contained a single set of genitalia in which medullary testes measured 0.09-0.13 (0.11 ± 0.01) mm long and 0.05-0.08 (0.06 ± 0.01) mm wide; a bi-lobed ovary was situated near the posterior margin of the proglottid, extending laterally up to the longitudinal excretory canals; the tubular uterus arose from the ootype up to the anterior margin of the proglottid; and vitelline follicles were cortical. The greater portion of the gravid proglottid was occupied by a uterus filled with unoperculate and embryonated eggs. Chemical analysis confirmed that the concentrations of heavy metals (Zn, Cu, Mn, Cd, Ni and Pb) accumulated in P. clarias were higher than in fish tissues and values recommended by FAO/WHO, with the exception of Zn, which was found to be higher in fish kidneys than in the cestode. This supports the hypothesis that cestodes of fish can be regarded as useful bioindicators when evaluating the environmental pollution of aquatic ecosystems by heavy metals.
